Research Abstract

I am a Senior Physician in Medical Oncology in the Division of Breast Oncology at Dana-Farber Cancer Institute (DFCI), an Associate Physician in Medical Oncology at Brigham and Women’s Hospital, and an Assistant Professor of Medicine at Harvard Medical School. I also serve as Clinical Director of the Center for Cancer Therapeutic Innovation (Early Drug Development Program) at DFCI, where I lead early-phase clinical development efforts for patients with metastatic breast cancer. My research focuses on therapeutic resistance and the development of novel treatment strategies for breast cancer, and I have served as Principal Investigator of 21 phase I clinical trials at DFCI, including first-in-human, phase I/Ib, and investigator-sponsored studies evaluating the safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary antitumor activity of investigational agents. This experience includes PI leadership of two phase I studies in triple-negative breast cancer: an investigator-sponsored trial of onvansertib plus paclitaxel and a phase I study of XMT-1660, a novel B7-H4-directed antibody-drug conjugate with a monomethyl auristatin E payload. These experiences provide me with the clinical, scientific, and operational expertise necessary to serve as Principal Investigator of the proposed first-in-human phase I trial of a novel antibody-drug conjugate in patients with metastatic triple-negative breast cancer after progression on a prior topoisomerase I inhibitor ADC, a population with substantial unmet clinical need. In this role, I will oversee dose escalation, expansion cohorts, safety monitoring, and integration of translational endpoints informed by preclinical studies conducted at DFCI and through the NIH NExT Program.
